This is a tough one, Nanbane, but I got a nominee after a lengthy inner debate. My choice, considering all your judgement criteria is:
"Maalaipozhuthin Mayakkathilae Naan Kanavukandaen Thozhi" by the goddess of TFM, P.Susheela.

"Athisaya Raagam, Aananda Raagam..." from
APOORVA RAAGANGAL" had great lyrics, great music
and great singing (Kannadasan/MSV/KJY). In my
opinion, this is one of the greatest Tamil movie
songs ever recorded.

My vote would be Kangal Enge by PS from Karnan. Actually many songs of Karnan would qualify for this. Pretty difficult to make a choice isnt it ?

my vote is for "mandhil urudi vendum" from sindhu bhairavi. very powerful yet simple lyrics from the one and only mahakavi, soft melody by IR delivered by the inimitable KJJ. can sing with just sruthi and enjoy it immensely.

My vote goes to "Kanne Kalaimaane" from Moondram Pirai.
The haunting flute, KJY's great voice and the lyrics suit the mood to a T.
I mean can you dream of any other song for that situation?

This is a very hard one, It's taken me ages and I decided to choose 3 songs(I had 2 extra by IR, as I tirelessly listen to them too and many more that I won't mention here.) Hope it's OK to have many songs here.
*NEW: Uyirum Neeye Uravum Neeye - Pavithra, MD ARR - The reason being, the music for this song is absolutely terrific (as for the raga, it is a Harikamboji derivative, like Kamas, I can't quite place it), the choice of instruments and rhythm sections fall in place like hand in glove. Unni Krishnan's beautiful voice and expression bring out the terrific lyrics which explain the relationship of a mother and child.
*IN-BETWEEN: Mayanginen solla thayanginen unnai virumbinen uyire - MD IR - This is a song produced by wonderful selection by the Maestro. He chose the ragam CHARUKESI, which I love very much and has produced an excellently different perspective of it. Another two songs where he has done this are Endhan Nenjil from Kalaignan, in NALINAKAANTHI(here, he used this ragam for the first time and created a phenomenon of a song in it) and Vedham Nee Iniya Naadham Nee in GOWLA(wow, what a tint he gave to this rarely used ragam, with instrumentation and beauty to this extent)
*OLD: Anthi mayanguthadi assai peruguthadi kanthan vara kaanene, vanna mayile vanna mayile - MLV
This song captured my heart as soon as I heard it rendered by the great ML Vasanthakumaari. The song, set in Yeman Kalyaani is a classic, it is catchy and also is within the reigns of Carnatic music. I love it so much, that I have sung it in some concerts of mine. The lyrics are pure love, but to the Lord Murugan.

I'd say a song written by great
KANNADASAN.When MSV asked a song for the
tune 'Na.. Nana.. Na.. Nana' kannadasan expessed
his feeling in a cheap way as"Innada tune ithu.
Yeatrum.. erakam.. ethugai monai yeathuvaumae
illama ?".
For that MSV "If you can't make up a song for this tune then you're unfit for write lyrics for any tune" Kannadasan rose up angrily and gave this lyric in just few seconds which is really so
impressive and If you never hear this song I'd
suggest you to hear this. The song is ..
"Vaan Nila.. Nila ulla.. Uun Vaalibam Nila..
Thean Nial.." it goes like that.
